If we want to get really technical, the tests and calculators are actually determining a resting (RMR) vs. basal metabolic rate (BMR). It's very, very difficult to get an accurate BMR and typically this is only done in laboratory settings.
The RMR is close to the BMR, so we use it for all intents and purposes.

There are several formulas used to estimate BMR. Chronometer implements the Mifflin St Jeor equation.
